Abstract
Urease is a nickel-dependent metalloenzyme that catalyzes the hydrolysis of urea into a-mmonia (NH3) and carbon dioxide (CO2), significantly increasing soil nitrogen availability and acting as a bacterial virulence factor. Primarily found in plants (e.g., jack beans), fungi, and bacteria, it was the first enzyme crystallized. Key applications include agricultural urea fertilizer efficiency, medical diagnostics (e.g., H. pylori tests), and industrial applications.
Description
Definition: A high-molecular-weight metalloenzyme (EC 3.5.1.5) containing nickel ions, crucial for nitrogen metabolism.
History: It was the first enzyme to be purified and crystallized, achieved by James B. Sumner in 1926, proving enzymes are proteins.
Occurrence: Found in bacteria, fungi, algae, and plants (especially leguminous seeds like jack beans and soybeans).
Structure: Composed of multiple subunits (often oligomeric), with UreC acting as the primary functional subunit.
Function
Reaction: Urease catalyzes the hydrolysis of urea into a-mmonia and carbon dioxide:
(NH2)2CO + H20 - CO2 + 2NH3
Mechanism: It increases the rate of urea breakdown, releasing a-mmonia, which acts as a nitrogen source for microorganisms and plants.
pH Regulation: Because it produces a-mmonia, urease activity significantly increases the pH (alkalinizes) of its environment.
Pathogenesis: In bacteria (e.g., Helicobacter pylori), urease helps neutralize stomach acid, allowing the pathogen to colonize the gastric mucosa.
Application
Agriculture: Used to improve fertilizer efficiency by breaking down urea, though it can cause nitrogen loss through a-mmonia volatilization.
Medical Diagnostics: The "Rapid Urease Test" (RUT) detects H. pylori in gastric biopsies; if urease is present, a pH indicator changes color due to a-mmonia production.
Biotechnology & Environment: Used in biosensors to detect heavy metals (which inhibit urease) and in bioremediation, such as in concrete repairing via microbially induced carbonate precipitation (MICP).
Industrial: Application in treating wastewater containing urea.
